摘要: Background BAFF and APRIL are members of TNF superfamily. They play vital roles in the pathogenesis autoimmune diseases. BCMA, a receptor, shows higher affinity for than BAFF. Previous studies found that ligand binding specificity BCMA may be determined by sequence outside DxL motif. Objective Investigate contribution segment motif with ligands. Method In this study, conservative region was divided into two segments: BCMA1 (NEYFDSLLHACIPC), DXL BCMA2 (QLRCSSNTPPLT), Two peptides corresponding to segments were synthesized their ligands detected competitive ELISA. BCMA1-Fc fusion protein also constructed, purified analyzed indirect Results had no inhibiting effect on interaction BCMA-Fc BAFF, but, it inhibited 22.5% 15.2% mAPRIL respectively. The rates 91.7%, but 80.6% mAPRIL, suggesting without BCMA2, bound well less efficiently mAPRIL. Conclusion These results suggest an important role selectivity its
