摘要: Genomic selection in modern farming demands sufficient semen production young bulls. Factors affecting quality and capacity bulls are not well understood; DNA methylation, a complicated phenomenon sperm cells, is one such factors. In this study, fresh frozen-thawed samples from the same Norwegian Red (NR) at both 14 17 months of age were examined for chromatin integrity parameters, ATP content, viability, motility. Furthermore, reduced representation bisulfite libraries constructed according to two protocols, Ovation® RRBS Methyl-Seq System (Ovation method) previously optimized gel-free method sequenced study methylome samples. Sperm analyses indicated that concentration, total motility progressivity old NR significantly higher compared individuals age. The percentage fragmented cells decreased with increasing Libraries Ovation exhibited greater read loss shorter size following trimming. Downstream reads obtained revealed similar global methylation but differentially methylated regions (DMRs) between 14- majority identified DMRs hypomethylated Most (69%) less than 10% difference while only 1.5% exceeded 25% difference. Pathway analysis showed genes annotated having low differences (less 10%) 10 differences, could be associated important hormonal signaling function relevant pathways, respectively. current research shows parallel routine informative reproductive Although levels similar, varying can linked pathways.
