摘要: The initial stages of the interaction between host and Aspergillus fumigatus at alveolar surface human lung are critical in establishment aspergillosis. Using an vitro bilayer model alveolus, including both epithelium (human adenocarcinoma epithelial cell line, A549) endothelium pulmonary artery cells, HPAEC) on transwell membranes, it was possible to closely replicate vivo conditions. Two distinct sub-groups dendritic cells (DC), monocyte-derived DC (moDC) myeloid (mDC), were included examine immune responses fungal infection surface. RNA high quantity quality extracted from layers membrane allow gene expression analysis using tailored custom-made microarrays, containing probes for 117 immune-relevant genes. This microarray data indicated minimal induction A549 response germ tubes A. fumigatus. In contrast, addition system greatly increased number differentially expressed moDC exhibited genes CLEC7A, CD209 CCL18 absence compared mDC. presence fumigatus, subgroups up-regulation identified previous studies as being associated with exposure exhibiting chemotactic properties neutrophils, CXCL2, CXCL5, CCL20, IL1B. approximated alveolus allowing pathogen interface that complements existing animal models IA.
