摘要: Apolipoprotein E (APO-E) binds to the beta-amyloid peptide and is present in senile neuritic plaques Alzheimer's disease (AD). The epsilon 4 isoform of APO-E has been associated with both sporadic familial late-onset AD, implying a causal role. Among patients control subjects similar age, gender, ethnic group from New York City community Washington Heights-Inwood, we found that odds ratio (OR) for AD homozygosity APO-epsilon was 17.9 (95% confidence interval [CI], 4.6-69.8) heterozygosity 4.2 CI, 1.8-9.5) compared persons other genotypes. association stronger among (OR = 10.3; 95% 3.4-31.1) than those family history dementia first-degree relative 0.9; 0.1-13.5). between did not differ according age at onset ( or 65), but appeared vary across 3 groups investigated (black, Hispanic, white). Our data confirm support hypothesis allele either confers genetic susceptibility may be linkage disequilibrium another locus. Ethnic variability allelic frequency elderly warrants further investigation.
