Mutational pattern of influenza B viruses adapted to high growth replication in embryonated eggs

摘要: Improved replication of influenza viruses in embryonated chicken eggs (CE) permits increased vaccine production and availability. We investigated the growth properties B relation to specific mutations occurring after serial passage CE. In experiments yielding high variants B/Victoria/504/2000, predicted alter amino acid (AA) composition occurred only near receptor-binding pocket hemagglutinins (HA) no other genes. Two B/Victoria/504/2000 had same AA substitutions HA (R162M D196Y), but higher yield variant a third substitution (G141E), which also altered antigenic characteristics. experiment B/Hong Kong/330/2001, PB2 NP domains relate RNP formation function. Our results indicate that adaptation high-yield by CE requires few either internal or external Specific modifications genes combination could be used optimize create for substrates.
