Nucleation of polar actin filament assembly by a positively charged surface.

摘要: Polylysine-coated polystyrene beads can nucleate polar assembly of monomeric actin into filamentous form. This nucleation has been demonstrated by a combination biochemical and structural experiments. The polylysine-coated accelerate the rate as detected two different assays. Subsequent examination electron microscopy reveals numerous filaments similar length radiating from beads. ATP promotes this bead-induced acceleration assembly. Decoration with myosin fragment S1 shows that these all have same polarity, arrowhead pattern pointing toward bead. relevance system to in vitro mechanisms its usefulness other studies are discussed.
