摘要: Glucocorticoids (GCs) exert their biological and therapeutical actions through the GC receptor (GR), a ligand-dependent transcription factor. Synthetic derivatives are widely prescribed for treating numerous cutaneous inflammatory immune diseases due to great efficacy. However, chronic treatment with GCs produces adverse side-effects including skin atrophy, delayed wound healing, in certain cases, resistance. The mechanisms underlying therapeutic of GR have been extensively studied; contrast, role as modulator epidermal development homeostasis has received less attention. ubiquitous functional inactivation results defective formation although not fully characterized. use transcriptomic approaches both vitro vivo allowed identification genes that regulated by developing adult skin. A main goal understand biology is identify primary transcriptional targets well signaling pathways mediating action. Furthermore, it will be important decipher contribution different cellular compartments skin, keratinocytes interfollicular epidermis hair follicles, respective stem cell progenitors. Additionally, recent findings indicating acts true peripheral endocrine organ implies greater complexity than originally thought. local production other steroid hormones should considered function under homeostatic diseased conditions. Finally, studying take into account mineralocorticoid may also mediate and/or regulate either itself or combination GR. Addressing these issues help elucidate which Gr contributes establishment competent barrier implications context dermatological treatments based on GC-analogs.
