摘要: EaCIR1, a 371-bp Erianthus-specific satellite DNA sequence, was cloned from TaqI restricted genomic after agarose-gel electrophoresis. This sequence has 77% homology with 365-bp of Helictotrichon convolutum and 72% 353-bp tandem repeat Oryza sativa. PCR primers defined in the conserved regions these repetitive sequences were used to isolate other DNAs different representatives Saccharum complex: SoCIR1 officinarum, SrCIR1 robustum, SsCIR1 SsCIR2 spontaneum, MsCIR1 Miscanthus sinensis. EaCIR1 localized subtelomeric chromosomes by fluorescence situ hybridization. Southern hybridization experiments, using two this family as probes, illustrated contrasting species-specificity demonstrated existence similar elements sorghum maize.
