摘要: Multiple applications of proteomics in life and health science, pathology pharmacology, require handling size-limited cell tissue samples. During proteomic sample preparation, analyte loss these samples arises when standard procedures are used. Thus, specific considerations have to be taken into account for processing, that summarised under the term microproteomics (μPs). Microproteomic workflows include: sampling (e.g., flow cytometry, laser capture microdissection), preparation (possible disruption cells or pieces via lysis, protein extraction, digestion bottom-up approaches, clean-up) analysis (chromatographic electrophoretic separation, mass spectrometric measurements statistical/bioinformatic evaluation). All steps must optimised reach wide dynamic ranges high numbers identifications. Under optimal conditions, is adapted studied types nature, isolates enriches whole content, clean-up removes salts other interferences such as detergents chaotropes, identifies many analytes instrumental throughput sensitivity allow. In suggested review, we present discuss current state μP processing small number (cell μPs) microscopic regions (tissue μPs).
