摘要: Sleep homeostasis, the increase in sleep observed following loss, is one of defining criteria used to identify throughout animal kingdom. As a consequence, deprivation and restriction are powerful tools that commonly provide insight into function. Nonetheless, experiments inherently problematic stimulus itself may be cause changes physiology behavior. Accordingly, successful techniques should keep animals awake and, ideally, result robust rebound without also inducing large number unintended consequences. Here, we describe technique for Drosophila melanogaster. The Nullifying Apparatus (SNAP) administers every 10s induce negative geotaxis. Although predictable, SNAP effectively prevents >95% nighttime even flies with high drive. Importantly, subsequent homeostatic response very similar achieved using hand-deprivation. timing spacing stimuli can modified minimize loss thus examine non-specific effects on assess arousal thresholds. disruption better understand
