The Role of Interleukin-10 and Hyaluronan in Murine Fetal Fibroblast Function In Vitro: Implications for Recapitulating Fetal Regenerative Wound Healing

摘要: Background Mid-gestation fetal cutaneous wounds heal scarlessly and this has been attributed in part to abundant hyaluronan (HA) the extracellular matrix (ECM) a unique fibroblast phenotype. We recently reported novel role for interleukin 10 (IL-10) as regulator of HA synthesis ECM, well ability produce an HA-rich pericellular (PCM). hypothesized that IL-10-mediated was essential functional phenotype and, moreover, could be recapitulated adult fibroblasts via supplementation with IL-10 dependent process. Methodology/Principal Findings To evaluate differences profile, we compared metabolism (MTS assay), apoptosis (caspase-3 staining), migration (scratch wound assay) invasion (transwell between C57Bl/6J murine (E14.5) (8 weeks) fibroblasts. found have lower rates apoptosis, increased migrate invade fibroblasts, these effects were on synthase activity. Further, addition resulted HA-dependent manner. Conclusions/Significance Our data demonstrates mediated is fibroblasts' enhanced properties. Moreover, mechanism can recapitulate aspect suggesting regenerative healing.
