摘要: The complete sequence of P30, the major surface Ag protozoan parasite, Toxoplasma gondii, has been deduced through cloning and analysis its gene. Using polyclonal serum specific for we have isolated a P30 cDNA clone from lambda gt11 expression library derived tachyzoites T. gondii (RH strain). This produces beta-galactosidase fusion protein which reacts with several anti-P30 mAb. In addition, anti-serum raised to purified exclusively in whole cell lysate gondii. was used isolate near full-length molecules cosmid containing Sequence reveals single open reading frame coding capacity 34.7 kDa primary translation product (consistent apparent Mr on SDS-acrylamide gels) including presumptive hydrophobic signal sequence. also carboxy-terminal tail is predictive posttranslational cleavage modification glycolipid anchor. We identified 5' 3' ends mRNA transcript extremely abundant, 1500 nucleotides length, polyadenylated. gene copy contains no introns.
