Electrokinetically controlled real-time polymerase chain reaction in microchannel using Joule heating effect

摘要: In this work, a new method to control polymerase chain reaction (PCR) thermal cycling in microchannel using Joule heating effect was presented. generated internally by the current flow through buffer solution an electrokinetically-driven microfluidic chip without external heater component. Numerical simulations were developed and conducted determine parameters required achieve desired cycling. A PDMS-based PCR fabricated tested. The power consumption only 1.3 W for such heating-controlled chip. To demonstrate feasibility of cycling, DNA fragment Escherichia coli O157:H7 stx1 successfully amplified two-temperature TaqMan real-time reaction.
