Distinct Roles of GABA(A) and GABA(B) Receptors in Balancing and Terminating Persistent Cortical Activity

摘要: Cortical networks spontaneously fluctuate between persistently active Up states and quiescent Down states. The are maintained by recurrent excitation within local circuits, can be turned on off synaptic input. GABAergic inhibition is believed to important for stabilizing such persistent activity balancing the excitation, could have an additional role in terminating state. Here, we report that GABAA GABAB receptor-mediated distinct complementary roles activity. In a model of Up–Down expressed slices rat entorhinal cortex, receptor antagonist, gabazine (50–500 nm), concentration-dependently decreased state duration, eventually leading epileptiform bursts. contrast, CGP55845 (50 nm 1 μm), increased duration network activity, prevented stimulus-induced transitions.
