摘要: The size distribution of the 20 aminoacyl-tRNA synthetses from wild-type Chinese hamster ovary (CHO) cells and mutant cell line tsH1, containing a temperature-sensitive leucyl-tRNA synthetase, was determined by gel filtration. Nine synthetases, specific for arginine, aspartic acid, glutamic glutamine, isoleucine, leucine, lysine, methionine proline, which coeluted as high-Mr entities (Mr∼ 1.2 × 106), were further co-purified to yield multienzyme complex, polypeptide composition identical that previously complex rabbit liver. Immunoprecipitates obtained crude extracts tsH1 cells, using antibodies directed lysyl-tRNA or methionyl-tRNA synthetase components displayed same compositions purified thereby establishing heterotypic nature this complex. Although activity grown at permissive temperature, low compared wild-type, Mr 129000, corresponding enzyme, present in similar amounts occurred exclusively component Finally, we report attempts demonstrate phosphorylation cultured CHO, HeLa C3 unsuccessful.
