摘要: In the melon exotic accession PI 161375, gene cmv1, confers recessive resistance to Cucumber mosaic virus (CMV) strains of subgroup II. cmv1 prevents systemic infection by restricting bundle sheath cells and impeding viral loading phloem. Here we report fine mapping cloning cmv1. Screening an F2 population reduced region a 132 Kb interval that includes Vacuolar Protein Sorting 41 gene. CmVPS41 is conserved among plants, animals yeast required for post-Golgi vesicle trafficking towards vacuole. We have validated as gene responsible resistance, both generating CMV susceptible transgenic expressing allele in resistant cultivar characterizing TILLING mutants with susceptibility CMV. Finally, core collection 52 accessions allowed us identify single amino acid substitution (L348R) only polymorphism associated phenotype. first natural found be involved transport its cellular function suggests might use own
