摘要: This study is an investigation to experimentally confirm whether or not ultrasound (US) irradiation effective in enhancing the vivo gene expression of plasmid DNA tumor. Dextran was cationized by introducing spermine hydroxyl groups allow polyionically complex with a DNA. The dextran prepared additionally modified poly(ethylene glycol) (PEG) molecules which have active ester and methoxy at each terminal, obtain different percentages PEG introduced. Various dextrans without introduction were mixed LacZ form dextran-plasmid complexes. Electrophoretical examination revealed that complexed both PEG-introduced dextran, irrespective percentage, although higher N/P ratio needed for complexation latter. By zeta potential changed be positive. charge complexes became close 0 mV as their percentage introduced increased, molecular size about 250 nm, introduction. When intravenously injected mice carrying subcutaneous Meth-AR-1 fibrosarcoma mass subsequent US tumor percutaneously, plus enhanced level significantly high extent compared free irradiation. depended on time period timing Fluorescent microscopic studies localization observed tissue We conclude combined promising way target expression.
