Accumbal strychnine-sensitive glycine receptors: an access point for ethanol to the brain reward system.

摘要: Background: Ethanol (EtOH), like other drugs of abuse, increases extracellular dopamine (DA) levels in the nucleus accumbens (nAc) brain reward system, an effect that may be importance for alcohol addiction. How this DA increase is produced not fully understood, although previous studies from present laboratories indicate nicotinic acetylcholine receptors ventral tegmental area play important role mediating effect. Furthermore, activation these secondary to some priming by EtOH nAc. We recently demonstrated strychnine-sensitive glycine (GlyRs) are nAc and they involved regulating levels. Here we examine tentative accumbal GlyRs above-mentioned mechanism EtOH. Method: In vivo microdialysis (coupled high pressure liquid chromatography with electrochemical detection) reversed microdialysis, awake, freely moving adult male Wistar rats. Results: Local perfusion strychnine decreased per se completely prevented after both local systemic administration. Accumbal GlyR agonist instead increased a subpopulation rats EtOH-induced but all animals. Conclusion: The results suggest might constitute targets its mesolimbic DA-activating Gene polymorphism drug developmental focus on receptor population relation dependence warranted.
