摘要: Abstract Aim The aim of this study is to determine the anticancer effect sulfite on SH-SY5Y neuroblastoma cells in vitro conditions and elucidate underlying molecular mechanism explore its therapeutic activity. Main methods In study, cytotoxic effects were detected over time a dose dependent manner with IC50 doses ranging from 0.5 to10 mM. Genotoxic was shown by comet assay. as 5 mM. Expression profiles target genes related apoptosis cell cycle control determined quantitative RT-PCR. Protein changes western blot analysis. Key findings URG4/URGCP, CCND1, CCND2, CDK4, CDK6, E2F4 BCL-2 gene expression levels significantly reduced RB1, TP53, BAX, BID, CASP2, CASP3, CASP9 DIABLO expressions increased group cells. result may be inhibition at G1 phase down-regulating URG4/URGCP or CDK6 stimulating via intrinsic pathway. Sulfite suppressed invasion colony formation line using matrigel chamber assay, respectively. Significance It thought that demonstrates anticarcinogenesis activity affecting arrest, apoptosis, invasion, an effective agent for treatment single combination other agents.
