Interaction of rat renal glucocorticoid receptor with Hsp90 and Hsp70 upon stress provoked by mercury.

摘要: The influence of mercury on the association rat kidney glucocorticoid receptor (GR) with heat shock proteins Hsp90 and Hsp70 was investigated. GR heterocomplexes were immunopurified from renal cytosol rats administered different doses (1, 2 3 mg Hg kg−1 b.w.). A quantitative immunoblotting procedure applied to determine levels GR, two nucleocytoplasmic isoforms (constitutive Hsp73 inducible Hsp72) in cytosol, as well amounts these within immunoprecipitated by anti-GR antibody. Mercury found stimulate all examined Hsps. most prominent effect metal stimulation Hsp72 interaction GR. On other hand, administration led an increase level while cytosolic remained unaltered. These findings suggest that Hsps, at least Hsp70, might be ascribed changes affinity their rather than Hsp availability cytosol. Therefore, heterocomplex assembly seems a controlled process enabling chaperoning functioning concert physiological demands. Copyright © 2006 John Wiley & Sons, Ltd.
