All- trans retinoic acid up-regulates the human CD2AP gene expression through Sp1/Sp3 binding sites

摘要: All-trans retinoic acid (ATRA), an active metabolite of vitamin A, plays important role in regulating cell differentiation, proliferation, and apoptosis. It was reported that ATRA could cause up-regulation protein expression CD2AP nephrotic animals. However, the mechanism ATRA-mediated is not well understood. In present study, deletion analysis luciferase assays demonstrated caused a marked increase activity promoter, region between nt −599 −328 from transcription start site, where there are two clusters Sp1/3 binding sites, indispensable for up-regulation. Chromatin immunoprecipitation revealed activated through enhancing DNA-binding Sp1 Sp3 with promoter. Taken together, this study provided evidence first time showing stimulating effect on new therapeutic strategies treatment nephritic syndrome other associated diseases deficiency.
