Canine distemper virus induces apoptosis in cervical tumor derived cell lines

摘要: Apoptosis can be induced or inhibited by viral proteins, it form part of the host defense against virus infection, a mechanism for spread to neighboring cells. Canine distemper (CDV) induces apoptotic cells in lymphoid tissues and cerebellum dogs naturally infected. CDV also produces cytopathologic effect, leading apoptosis Vero tissue culture. We tested canine virus, member Paramyxoviridae family, ability trigger HeLa cells, derived from cervical cancer resistant apoptosis. To study effect infection we examined markers 24 h post (pi), flow cytometry assay DNA fragmentation, real-time PCR caspase-3 caspase-8 mRNA expression, -8 immunocytochemistry. Flow showed that fragmentation was infected CDV, immunocytochemistry revealed significant increase levels cleaved active protein, but did not show any difference expression caspase-8, indicating an intrinsic pathway. Confirming this observation, higher than control cells; however, there no statistically change profile. Our data suggest triggering pathway, with participation initiator caspase extrinsic In conclusion, cellular stress caused apoptosis, used as tool future research treatment control.
