Intracellular colonization of roots of Arabidopsis and crop plants by Gluconacetobacter diazotrophicus

摘要: We have investigated the interaction of Gluconacetobacter diazotrophicus, a non-nodulating endophytic nitrogen-fixing bacterium isolated from intercellular spaces sugarcane, with Arabidopsis thaliana and crop plants maize (Zea mays), rice (Oryza sativa), wheat (Triticum aestivum), oilseed rape (Brassica napus), tomato (Lycopersicon esculentum), white clover (Trifolium repens). Using seedlings grown aseptically in sucrose-containing culture media, we shown that inoculation very low numbers G. diazotrophicus results extensive intracellular colonization root meristems progressive systemic colonization. Light microscopic examination thin sections resin-embedded tips these inoculated β-glucuronidase (GUS)-labeled NifH promoter-GUS-labeled showed blue-stained within cytoplasm cells, indicating conditions were suitable for nitrogenase gene expression. Electron microscopy confirmed bluestained membrane-bounded vesicles. discuss whether novel inoculations are likely to enable non-nodular endosymbiotic nitrogen fixation can also provide plant system investigate theory origin eukaryotic organelles.
