Instability of long inverted repeats within mouse transgenes.

摘要: Various sequences in the mammalian genomes are unstable. One class of sequence arrangement is long inverted repeats, which known to be unstable bacteria and yeast. While mammals some evidence suggests that short repeats (<10 bp long) may show instability, nothing about stability repeats. Here we describe two unrelated multicopy transgenes mouse (loci 109 OX1-5), each contains a repeat shows substantial mitotic instability. This instability also occurs germline so mutant appear within pedigrees at high frequency. The mutation processes acting these complex can involve insertion or deletion, result stabilization transgene. At transgene mutational events range from very small rearrangements centre complete deletion. In addition rates OX1-5 vary between male female germlines inbred strains mice, suggesting possibility genetic analysis identify loci modulate
