Quantification of the cell kinetic effects of G-CSF using a model of human granulopoiesis

摘要: Using a mathematical model of normal human granulopoiesis, the most important influences granulocyte colony-stimulating factor (G-CSF) on regulation white cell production can be quantified. G-CSF increases blood neutrophils via three effects: reduction transit time postmitotic granulopoietic bone marrow cells; additional mitoses early and possibly demargination. The application in vivo results summation single effects. For 5 days at daily dose 10 micrograms/kg, from 6 to 1.5 together with demargination rate 10% per hour explains initial increase until day 1. Both effects fail explain sustained thereafter. Enhanced amplification cells is also necessary reproduce increase. Depending individual clinical data, approximately occur, evenly distributed myeloblasts, promyelocytes myelocytes. If above are assumed, predicts data accordance observations.
