摘要: This study was aimed to evaluate the synergistic effects of polyamines (PAs) and plant growth regulators (PGRs) on in vitro propagation stevioside production Stevia rebaudiana (Stevia). A large-scale protocol established for leaf explants Murashige Skoog medium (MS) supplemented with various combinations PAs PGRs. The combination spermidine (Spd, 2.0 mg L−1) 2, 4-dichlorophenoxy acetic acid (2, 4-D, 1.5 mg L−1) 6-benzyleadenine (BA, 1.5 mg L-1) induced maximum callogenic response (91.7%). Spd (1.0 mg L−1) BA found most effective shoot regeneration (94.4%), mean number shoots (14. 7), leaves per explant (88.3). However, putrescine (Put, kinetin (Kn, promoted length (6.6 cm). Incorporation either or Put naphthalene (NAA) indole butyric (IBA) culture media improved root organogenesis. Vigorous plantlets having optimum roots were successfully acclimatized soil. Chromatographic data revealed that synergism Spd, BA, Kn (2.0 mg L−1) enhanced content (10.20 mg/g DSB) as compared control (3.02 mg/g DW). Furthermore, application fresh (57.5 g L−1 FSB) dry biomass (9.03 g L−1 control. In contrast, increased antioxidant activity (80.6%) (55.3%). Combination GA3 (2.0 mg phenolic flavonoid contents. is first successful report accumulation higher content, a potential step towards industrial production.
