摘要: The role of myelin proteolipid apoprotein (PLP) in the central nervous system (CNS) immune response rabbits has been investigated by analyzing immunopathology chronic experimental allergic encephalomyelitis (EAE) induced sensitization with PLP. Clinical disease occurred seven out nine sensitized bovine PLP and monitored for up to 6 mo. Positive delayed hypersensitivity skin test reactions all but one PLP-sensitized animals. All animals had meningeal CNS parenchymal inflammation that correlated severity. Serial blood samples were stained a panel antibodies rabbit T B cells, as well Ia, large small mononuclear cell populations analyzed flow cytometry. Peripheral leukocyte population staining did not correlate clinical signs or Cryostat tissue sections same set using an immunoperoxidase technique, positive cells vessels counted. macrophages numerous equal numbers perivascular inflammatory infiltrates, whereas less (p than 0.001). also diffusely infiltrated parenchyma. Most many scattered Ia+; Ia vascular expression was increased over controls 0.001), this EAE model is whole tissue- basic protein-induced other species, similar multiple sclerosis. Cellular responses may therefore contribute systemic situ demyelinating diseases.
