摘要: Summary. This study compares the utility of two functional assays for von Willebrand factor (VWF), ristocetin cofactor assay (VWF:RCo) and collagen-binding (VWF:CBA). We analysed a group 32 patients with type 2 disease (VWD) (25 2M, six 2A one 2B) 22 normal control subjects. VWF:RCo/VWF antigen (VWF:Ag) ratios VWF:CBA/VWF:Ag were compared between patient groups. In VWD, both VWF:RCo/VWF:Ag discordant (≤ 0·7). 25 2M VWD patients, concordant (> 0·7), but VWF:RCo/VWF:CBA (≤ 0·7) (P = 0·001) Thus, in groups indicating abnormality. However, not group. Our showed that VWF:CBA is sensitive to variants associated loss high-molecular-weight multimers, i.e. 2B was unable discriminate defective platelet-binding multimeric patterns such as VWD. It concluded should be used association rather than replacement VWF:RCo assay.
