摘要: Background: The genus Populus is accepted as a model system for molecular tree biology. To investigate gene functions in spp. trees, generating stable transgenic lines the common technique functional genetic studies. However, limited number of genes have been targeted due to lengthy process. Transient transformation assays complementing significant advantages rapid vivo assessment function. aim this study develop simple and efficient transient hybrid aspen provide its potential applications genomic approaches. Results: We developed an planta assay young cuttings using Agrobacterium-mediated vacuum infiltration. conditions such infiltration medium, presence surfactant, phase bacterial growth density were optimized achieve higher efficiency leaves. Agrobacterium successfully transformed various cell types leaf tissues. Intracellular localization four was confirmed homologous fusion constructs with green fluorescent protein. Protein-protein interaction detected transiently co-transformed cells bimolecular fluorescence complementation technique. In promoter activity monitored over few days that luciferase reporter driven by circadian clock promoter. Conclusions: here enhanced throughput method requires minimum handling short This will facilitate analyses plant system.
