摘要: The RIG-I-like receptors family is a group of cytosolic RNA helicase proteins that can recognize viral via binding to pathogen associated molecular pattern motifs within ligands. A novel vertebrate RLR counterpart named LjMDA5 was firstly identified from the marine fish sea perch Lateolabrax japonicus in this study. full-length cDNA 3750 bp and encodes polypeptide 988 amino acids, containing two N-terminal tandem caspase activation recruitment domains, DExH (Asp-Glu-X-His) box domain, an HELICc C-terminal domain RIG-I. Phylogenetic analysis showed shared closest genetic relationship with MDA5 Larimichthys crocea. Quantitative RT-PCR ubiquitously expressed up-regulated significantly all selected tissues vivo post NNV infection. Time course transcripts increased spleen kidney. We found could be regulated LJB LJF cell lines after lipopolysaccharide, polyinosinic-polycytidylic acid treatment challenge. interference experiment indicated silencing RGNNV replication virus production infected cells. Our results revealed essential for host defense against NNV, which provided new insights into function signaling pathway during infection fish.
