摘要: Pasteurella haemolyticaleukotoxin (LKT) is a member of the RTX family pore-forming toxins that kill bovine immune cells. Several studies have suggested target cells by induction apoptosis. In present study, BL3 leukaemia were exposed to LKT and assessed molecular flow cytometric techniques measure different aspects apoptotic cell death. The intoxicated demonstrated morphological, light scatter Hoechst 33258 staining characteristics consistent with undergoing also exhibited internucleosomal DNA fragmentation poly (ADP-ribose) polymerase (PARP) cleavage, both indicators LKT-treated bound annexin-V-FITC indicating phosphatidylserine groups translocated from inner outer leaflet membrane. effect on was dose dependent inhibitable incubation anti-LKT monoclonal antibody. Finally, an early step for apoptosis appears be binding β2 integrin since pre-incubating anti-β2 antibodies inhibited LKT-induced This study provides new insights into understanding pathogenesis pasteurellosis could lead development preventative therapeutic strategies disease management.
