摘要: Objective: A simple method for accounting efficiently multiple testing of many SNPs in an association study was recently proposed by Nyholt, but its performance not extensively evaluated. The involves estimating ‘effective number’ independent tests and then adjusting the smallest observed p value using Sidak’s formula based on this number tests. We sought to carry out empirical theoretical evaluation Nyholt’s method. Methods: applied a sample 31 genes typed at total 291 permutation used determine type-I error rate each gene. Based our results, we algebraically investigated effective model haplotype block structure. Results: nominal 5% type I varied from under 3% over 7%, dependent linkage disequilibrium. Theoretical considerations show further that can be very conservative presence Conclusion: Although approach may useful as exploratory tool, it is adequate substitute
