Differential effects of two lots of aroclor 1254: congener-specific analysis and neurochemical end points.

摘要: Aroclor 1254 is a widely studied commercial polychlorinated biphenyl (PCB) mixture which, by definition, contains 54% chlorine weight. Recent reports indicate substantial differences in the congener composition among lots and hence their biologic effects. We designed current study to compare effects of two (lots 6024 124-191). analyzed these for PCB congeners, dibenzofurans (PCDFs), naphthalenes (PCNs), dibenzodioxins (PCDDs). used previously established techniques analyzing intracellular Ca(2+) buffering protein kinase C (PKC) translocation test activity neuronal preparations. congener-specific analysis indicated that ortho non-ortho congeners varied percent contribution. Among all percentages (PCBs 77, 81, 126, 169) were higher lot (2.9% total) than 124-191 (0.02% total). detected no dioxins (< 2 ppb). Although there are some composition, total PCNs similar both lots: 171 ppm 155 124-191. However, PCDFs (38.7 ppm) (11.3 ppm). When we tested Aroclors on PKC brain preparations, significantly different. was more potent 6024, slightly active These could not be attributed percentage or because they inactive parameters. The levels almost similar. seen with cells also predicted based TCDD toxic equivalents (TEQs), although TEQs ethoxyresorufin-O-deethylase (EROD) methoxyresorufin-O-deethylase (MROD) activities. It possible differential caused ortho-congeners mixtures, PCBs ortho-lateral substitutions can exhibit different activities selected neurochemical end points.
