摘要: In this study, DNA constructs containing the putative zebrafish promoter sequences of GATA-1, an erythroid-specific transcription factor, and green fluorescent protein reporter gene, were microinjected into single-cell embryos. Erythroid-specific activity GATA-1 was observed in living embryos during early development. Fluorescent circulating blood cells detected 24 hours after fertilization still present 2-month-old fish. Germline transgenic fish obtained from injected founders continued to express erythroid F1 F2 generations. The expression patterns consistent with pattern mRNA by RNA situ hybridization. These have allowed us isolate, fluorescence-activated cell sorting, earliest progenitor developing for vitro studies. By generating using other promoters it should be possible visualize origin migration any lineage-specific a embryo.
