摘要: Janus kinases (Jaks) are a family of nonreceptor tyrosine kinase that include four different members, viz., Jak1, Jak2, Jak3, and Tyk2. Jaks play critical roles in immune cells functions; however, recent studies suggest they also essential nonimmune cell physiology. This review highlights the significance epithelial understanding molecular basis some diseases through regulation epithelial-mesenchymal transition, survival, growth, development, differentiation. Growth factors cytokines produced by hematopoietic origin use Jak for signal transduction both cells. Among Jaks, Jak3 is widely expressed intestinal (IECs) humans mice. Mutations abrogate functions cause an autosomal severe combined immunodeficiency disease (SCID) while activating mutations lead to development hematologic cancers. A selective inhibitor CP-690550 (Xeljanz) approved FDA certain chronic inflammatory conditions demonstrates immunosuppressive activity rheumatoid arthritis, psoriasis, organ transplant rejection. Here, we focus on consequences Jak3-directed drugs adverse effects light discoveries mucosal with information other Jaks. Lastly, brief structural implications domains beyond As about gastrointestinal associated only just emerging, review, summarize its wound repair, bowel disease, obesity-associated metabolic syndrome, shed lights identifying potential novel targets developing therapeutic interventions dysfunctional IEC.
