摘要: The development of a clear understanding the physiology marine prokaryotes is complicated by difficulties inherent in resolving activity various components natural microbial communities. Application appropriate molecular biological techniques offers means overcoming some these problems. In this regard, we have used direct probing bulk RNA purified from selective size fractions to examine variations rRNA content heterotrophic communities and Synechococcus populations on southeastern U.S. continental shelf. Heterotrophic seawater cultures amended with selected substrates were examined. isolated water column differential filtration. total cellular target was assayed samples an oligonucleotide complementing universally conserved region eubacterial 16S (heterotrophs) or 1.5-kbp fragment encoding sp. strain WH 7803 (cyanobacteria). analyses revealed that bacteria responded addition glucose trace nutrients after 6-h lag period. However, no response detected amino acids added. increased 48-fold before dropping value 20 times Variations spp. followed distinct diel pattern imposed phasing cell division within irradiance cycle. results indicate careful application can be great use discerning basic physiological characteristics mechanisms which regulate growth at subcellular level.
