摘要: Abstract Sulfonated steroids (s-St) have been usually regarded as inactive metabolites but are progressively considered precursors for the intra-tissue formation of bioactive steroids. Moreover, independent effects without preceding removal sulfate group observed. We use porcine testicular-epididymal compartment a model to investigate still largely unknown s-St physiology boar exhibits an intriguingly broad spectrum predominantly originating from testis. The application LC–MS/MS in steroidomics enables determination unconjugated and intact sulfonated with currently highest specificity good sensitivity, allowing concurrent measuring numerous analytes larger quantities samples. Profiles (6 h, 20 min intervals) were generated 5-androstene-3s,17s-diol (Adiol-S), androsterone (A-S), dehydroepiandrosterone (DHEA-S), epiandrosterone (EA-S), epitestosterone (ET-S), estrone (E1-S), estradiol-17β (E2-S), pregnenolone (P5-S), 17αOH-pregnenolone (OHP5-S) testosterone (T) four unstimulated hCG-stimulated boars. concentrations measured individual samples collected testicular afferent efferent blood differentiate between vs. extratesticular origin. Highest found EA-S, followed by ET-S, Adiol-S DHEA-S, which mostly exceeded levels E1-S A-S. Lowest obtained E2-S, P5-S OHP5-S. analytical profile also included T, 5α-dihydrotestosterone cholesterol. However, their below limit quantification. quantifiable consistent wave-like pattern associated T pulses. In postpartal females (n = 5) all assessed undetectable, suggesting that pigs adrenals not quantitatively significant source s-St.
