Metabotropic Glutamate Receptor Subtype 8 in the Amygdala Modulates Thermal Threshold, Neurotransmitter Release, and Rostral Ventromedial Medulla Cell Activity in Inflammatory Pain

摘要: The amygdala is a crucial area in controlling the threshold of pain and its emotional component. present study has evaluated effect metabotropic glutamate 8 receptor (mGluR8) stimulation central nucleus (CeA) on thermoceptive CeA serotonin (5-HT), (Glu), GABA release normal carrageenan-induced inflammatory conditions rats. Furthermore, activity rostral ventromedial medulla (RVM) putative “pronociceptive” ON “antinociceptive” OFF cells been evaluated. (S)-3,4-Dicarboxyphenylglycine [(S)-3,4-DCPG], selective mGluR8 agonist, administered into CeA, did not change 5-HT, Glu, release, or threshold, nor it modify RVM animals. In rats treated with carrageenan, intra-CeA (S)-3,4-DCPG perfusion produced antinociception, increased 5-HT whereas decreased release. Intra-CeA inhibited cell activities. an increase gene, protein, staining, latter being associated vesicular transporter-positive profiles, found after pain. These results show that mGluR8, which was overexpressed within conditions, inhibits nociceptive behavior. Such Glu decrease GABA, inhibition ON- OFF-cell activities RVM.
