Multiplex assays for biomarker research and clinical application: Translational science coming of age

摘要: Over the last decade, translational science has come into focus of academic medicine, and significant intellectual financial efforts have been made to initiate a multitude bench-to-bedside projects. The quest for suitable biomarkers that will significantly change clinical practice become one biggest challenges in medicine. Quantitative measurement proteins is critical step biomarker discovery. Assessing large number potential protein statistically samples controls still constitutes major technical hurdle. Multiplexed analysis offers advantages regarding time, reagent cost, sample requirements amount data can be generated. two contemporary approaches multiplexed quantitative validation, antibody-based immunoassays MS-based multiple (or selected) reaction monitoring, are based on different assay principles instrument requirements. Both their own disadvantages therefore complementary roles multi-staged verification validation process. In this review, we discuss immunoassay monitoring/selected monitoring development. We also choosing an appropriate platform, judging performance assays, obtaining reliable, results research applications field.
