摘要: A method for estimating and comparing population genetic variation using random amplified polymorphic DNA (RAPD) profiling is presented. An analysis of molecular variance (AMOVA) extended to accomodate phenotypic data in diploid populations Hardy-Weinberg equilibrium or with an assumed degree selfing. We present a two step strategy: 1) Estimate RAPD site frequencies without preliminary assumptions on the unknown structure, then perform significance testing substructuring. 2) If structure evident from first step, use this calculate better estimates sub-population components. nonparametric test homogeneity (HOMOVA) also This was designed statistically differences intrapopulational variances (heteroscedasticity among populations). These theoretical developments are applied set Vaccinium macrocarpon (American cranberry) small sample sizes, where gradient diversity found between central marginal populations. The AMOVA HOMOVA methods provide flexible tools when other that many markers direct allelic data.
