Nucleotide composition and kinetic complexity of the genomic DNA of an intracellular symbiont in the pea aphid Acyrthosiphon pisum

摘要: An intracellular symbiont was isolated from the mycetocyte of pea aphidAcyrthosiphon pisum, and its genomic DNA compared with those ofEscherichia coli andMycoplasma capricolum respect to nucleotide composition kinetic complexity. Thermal dissociation, CsCl density equilibrium centrifugation, high-performance liquid chromatography nuclease P1 digest all indicated that G+C content endosymbiont is as low 30%. In this respect, resembledMycoplasma species. The reassociation kinetics labeled by nick translation suggested genome 1.4×1010 daltons in size, about 5 18 times large ofE. andM. capricolum, respectively. results were confirmed incubation [3Hthymidine Grace's medium. aphid 500 larger than leafhopper endosymbionts previously analyzed ultracentrifugation.
