An innovative sandwich ELISA system based on an antibody cocktail for gluten analysis

摘要: A cocktail sandwich ELISA based on the employ of two monoclonal antibodies (MAbs) as coating and a third MAb conjugated to horseradish peroxidase has been developed for analysis gluten in foods. Given that each displays wide specificity spectrum wheat, barley, rye oats prolamins, their combination ensures high crossreactivity with most potentially toxic gliadin, hordein, secalin avenin protein family. One unprecedented features is it permits first time barley hordeins foods, which unattainable using conventional or commercial kits. Besides, gliadins, secalins are recognised same extent. The system provides detection sensitivity hordeins, avenins (1.5, 0.05, 0.15 12 ng/ml, respectively). working linear range comprises 3–100 ng/ml gliadin limit 1.5 ppm. This even better than demanded latest Codex recommendation, 10 Cocktail data were contrasted those kits confirmed by mass spectrometry, non-immunological technique evidence occurrence false positive results
