Sister-chromatid exchanges in human lymphocytes exposed to 1-p-(3-methyltriazeno)benzoic acid potassium salt.

摘要: The 1-p-(3-methyltriazeno) benzoic acid potassium salt (MTBA) is a triazeno analogue of dacarbazine, an antineoplastic agent capable mediating the appearance new antigenic specificities on cancer cells in mice, phenomenon described as 'chemical xenogenization' (CX). Recently we reported clastogenic potential MTBA human lymphocytes. Since sister-chromatid exchange (SCE) assay more sensitive than tests, at least low drug concentrations, assessed SCE frequencies induced by lymphocytes stimulated PHA. Drug treatment 2-500 micrograms/ml was performed vitro prior to or after PHA addition. values increased significantly dose-dependent manner up 200 micrograms/ml. However, frequencies, well chromosome breaks, did not increase dramatically. These data indicate that concentrations used for CX do cause severe cytogenetic damage immune vitro.
