摘要: AIM: To investigate the transactivating effect of HCV core protein on Wee1 gene and molecular biological mechanisms in pathogenicity. METHODS: Polymerase chain reaction (PCR) technique was employed to amplify sequence promoter from HepG2 genomic DNA, product cloned into pGEM-T vector. The Weep cut T-Weep by KpnI XhoI, then pCAT3 basic, named pCAT3- Weep. pCAT3-Weep transfected hepatoblastoma cell line cotransfected cells with pcDNA3.1(-)-core FuGENE 6 transfection reagents. pCAT3-basic used as negative control. activity CAT detected an ELISA kit after 48 hours, which reflected function promoter.
