Alteration in Regulation of Serotonin Release in Rat Dorsal Raphe Nucleus after Prolonged Exposure to Morphine

摘要: Regulation of serotonin (5-HT) release may be altered during the development opioid tolerance and dependency. To test this hypothesis, changes in extracellular 5-HT prolonged administration morphine were determined by microdialysis dorsal raphe nucleus (DRN) freely behaving rats. Morphine or placebo pellets implanted s.c. As compared to placebo, induced a sustained, ∼50% increase DRN significant elevation hot plate latency 12-hr period after implantation. One week later had returned control levels, implanting additional no effect on latency. day removing from rats exposed for 2 wk, acute challenge with (20 mg/kg, s.c.) significantly smaller as treatment group. Administration naltrexone wk signs withdrawal decrease 5-HT. These results suggest that regulation is morphine. Thus, one factors involved physiology behavioral state withdrawal.
