Production of 10-Ketostearic Acid from Oleic Acid by Flavobacterium sp. Strain DS5 (NRRL B-14859).

摘要: A microbial isolate, Flavobacterium sp. strain DS5, produces 10-ketostearic acid (10-KSA) from oleic in 85% yield. This is the first report on this type of reaction catalyzed by a strain. The product was purified to give white, plate-like crystals melting at 79.2°C. optimum time, pH, and temperature for production 10-KSA are 36 h, 7.5, 30°C, respectively. small amount 10-hydroxystearic (10-HSA) (about 10% main product, 10-KSA) also produced during bioconversion. not further metabolized DS5 accumulates medium. In contrast growing cells, resting-cell suspension 10-HSA ratio 1:3. crude cell extract obtained ultrasonic disruption cells converts mainly (10-HSA/10-KSA ratio, 97:3). result strongly suggested that converted via 10-HSA. Enzymes catalyzing hydration secondary alcohol dehydrogenation associated. Product 66% enantiomeric excess 10(R) form. conversion enzyme(s) reacts with unsaturated fatty acids order > palmitoleic linoleic linolenic γ-linolenic myristoleic acid.
