摘要: Abstract Ethnopharmacological relevance Astragaloside IV (As IV) is one of the main effective components isolated from traditional Chinese medical herb Astragalus membranaceus. The protective effect membranaceus on myocardial hypertrophy has been extensively proved. To test hypothesis that can ameliorate and inflammatory induced by β-adrenergic hyperactivity, we carried out in vivo vitro experiments. Material methods In study, isoproterenol (Iso) (5 mg kg−1 d−1) was used as a model intraperitoneal injection. SD rats were randomly assigned to following six groups: A: control; B: Iso group; C: plus As 20 mg kg−1 d−1; D: 40 mg kg−1 d−1; E: 80 mg kg−1 d−1; F: Propranolol 40 mg kg−1 d−1. cultured neonatal rat cardiomyocytes pretreated with (3, 10, 30 μ mol L−1), (2 μ mol L−1) BAY11-7082 (5 μ mol L−1) for 30 min, then incubated (10 μ mol L−1) 48 h. For each group, heart mass index (HMI) left ventricular (LVMI) measured. measure transverse diameter cells (TDM), hematoxylin-eosin (HE) staining method applied. addition, volume total protein content measured, mRNA expression ANP TLR4 quantified RT-PCR, TLR4, IκBα p65 Western blot, level TNF-α IL-6 measured ELISA. Results vivo: Comparing group control, HMI, LVMI, TDM significantly increased; increased, while decreased; ANP, mRNA, TNF-α, serum increased. These changes could be partly prevented Pro. vitro: over-expression cell size, remarkably down-regulated Pro, results blot ELISA similar those vivo. Conclusions these studies indicate good isoproterenol. More specifically, cardioprotection related inhibiting TLR4/NF-кB signaling pathway attenuating effect.
