摘要: We have developed and tested a novel electrofusion chamber, the adjustable plate microchamber, that permits successful production of hybridomas in hanging droplet from as few 1,000 B lymphocytes. Cell suspension volumes 10 microliters may be used without excessive difficulty aseptically recovering fused cells. With modification hypo-osmolar protocol with this fusion efficiencies order 10(-3) attained. These are comparable to those attained standard hardware much higher numbers input This technology should permit high efficiency hybridoma using variety hitherto inaccessible lymphocyte populations such cells isolated human organ biopsies.
