摘要: PROBLEM: Major histocompatibility complex (MHC) class II molecule expression is specifically suppressed on fetal trophoblasts, even in response to interferon (IFN)-γ, a potent inducer of MHC genes. The suppression induction has been suggested play role preventing rejection the allograft. mechanism this unknown. METHOD OF STUDY: Human trophoblast cell lines were examined for transcription factors and activity IFN-γ signaling pathway. Additionally, cells transfected with vector expressing transactivator, CIITA, assayed expression. RESULTS: RFX X2BP pathway components are expressed constitutively functional trophoblasts. However, CIITA was absent trophoblasts could not be induced by IFN-γ. Transfection into resulted derepression gene expression. CONCLUSIONS: lack genes caused neither absence that bind promoters, nor lesion pathway, but results from specific inhibition gene.
