Single-chain intracellular antibodies inhibit influenza virus replication by disrupting interaction of proteins involved in viral replication and transcription

摘要: Influenza A virus is responsible for influenza epidemics in avian and human populations poses a great threat to health. Many researches have been focused on the prevention treatment of infection. The nucleoprotein (NP) an important protein due its ability interact with variety viral cellular factors role RNA synthesis. In this study, we used as target anti-viral therapy through new approach. By screening single-chain intracellular antibody (intrabody) cDNA library using bait yeast antigen-antibody two-hybrid system, identified several intrabodies that specifically nucleoprotein. Interaction between anti-nucleoprotein was further confirmed by mammalian analysis. Results showed changed their distribution association Further studies indicated abolished accumulation not only complementary virion but also messenger virus. addition, significantly inhibited transcription replication blocking interaction polymerase proteins, basic 1 2. Thus, study provides powerful tool protein's functions, opens potential avenue infections.
